Sterilize loop and let cool. spread aroundbacteria on slide. Let dry. Add several drops of95% methanolto the air-dried smear of bacteria andlet sit for2 minutes. Pick up the slide andallow the excess methanol to drain off. Let the slideagainair drybefore staining.Broth tube- draw nickel sized circle on bottom of slide with sharpie. Place 1-2 loops of bacteria incircle. Smear to thin film in circled area. All other steps the same.2. Define the following: acidic dye, basic dye, direct stain, and indirect stain.
3. State in chemical and physical terms the principle behind direct staining and the principlebehind indirect staining.(organisms have slight negative charge)
B. DIRECT STAININGRESULTS1. Recognize a direct stain preparation when it is observed through a microscope and statethe shape and arrangement of the organism.
