South 1 Maya South DNA Extraction Lab Purpose: In the lab you will extract DNA from green peas, strawberries or bananas. Hypothesis: If the banana contains DNA, it will appear as a white stringy solid substance between a water layer (that was left behind) and alcohol layer, which is DNA extracted in the form of a precipitate. Materials: bananas (1/2 cup of either one) a blender measuring cups measuring spoons a strainer small glass containers wooden stir stick table salt cold water liquid detergent pineapple juice isopropyl alcohol Procedure: 1. Add 100 ml of bananas, 1ml of salt and 200 ml of cold water into the blender. 2. Blend on high for 15 seconds 3. Pour the blender mixture through a strainer into a measuring cup 4. Add 30 ml (2 Tablespoons) of liquid detergent to the mixture, swirl and let sit for 7 -8 minutes 5. Pour the mixture into several small glass containers until about a third full 6. Add a small splash (⅛ tsp) of pineapple juice into each container 7.
