Reverse TranscriptionPolymerase ChainReaction (RT-PCR)
DefinitionReverse transcription polymerase chain reactionis a laboratory technique combining reversetranscription of RNA into DNA and amplificationof specific DNA targets using polymerase chainreaction. It is primarily used to measure theamount of a specific RNA.
•Reverse transcription polymerase chain reaction (RT-PCR)is one of many variants of polymerase chain reaction (PCR).•This technique is commonly used in molecular biology todetect RNA expression.•RT-PCR is often confused with real-time polymerase chainreaction (qPCR). However, they are separate and distincttechniques.•RT-PCR is used to qualitatively detect gene expressionthrough creation of complementary DNA (cDNA) transcriptsfrom RNA.
PrincipleThe principle is to convert RNA into itscomplementary DNA sequence by reversetranscriptase, tosynthesiseasecondstrand with DNA polymerase, and finally togenerate a cDNA molecule which can beamplified by PCR in the normal way
