Q: Is it OK to assume that all proteins in the Eluate 1 and 2 are YqhD to calculate the amount of YqhD? Based on our SDS-PAGE result, its not 100% YqhD, but I don’t know how to quantify the % purity of YqhD. A: Yes, you need to assume that all the protein in these elution fractions is YqhD. Q: For the table on pg. 37 of the lab manual, does the "X" for purified YqhD represent either 5 ul or 10 ul? Since we are performing the assay using 5 ul and 10 ul, we are trying to find the amount of enzyme that generates a linear decrease in absorbance at 340nm. Once we find the optimal enzyme amount, do we consider this amount as "X"? A: Yes, you should determine in 6 A, if μL or 10 μL work better and then use this for the assays in 6 B and C. Q: I am currently trying to figure out the distances that the standards have travelled on my gel but it is really difficult to distinguish some of the bands. Should I attempt to include values for all the bands in my standard curve? Also, the final two rows (not including the very last row) was my eluate 1 and 2. It
