The purpose of a stacking gel is to allow proteins to migrate through more quickly and more uniformly, since a large pore gel has less of a sieving effect. As a result the proteins are condensed into a tight band before entering the running gel. This allows for a sharper resolution of proteins within the running gel. 2. Why do we add SDS to our samples? We add SDS to our samples because when protein molecules unfold the SDS coats the unfolded proteins and prevents the protein molecule from refolding and masks the charges of the protein molecules. SDS allow proteins to have same streamlined shape and have their charges masked. The proteins have the same shape / and charge density. This allows proteins to differ from one another only in size.
