Examining Cross-over and Hybridization through Mapping of Sordaria Chromosomes Simon Alexander, March 3, 2008, Ridgway Abstract The fungus Sordaria fimicola was used to understand Mendel’s law of segregation, and to determine the frequency of cross-over between two different strains of wild-type (normal) and mutant gene of the Sordaria fungi. In order to identify the hybrids that had occurred after the period of incubation, we examined the physical appearance of the asci to find out how much cross-over between the fungi had taken place. After we had calculated the map units for the given data we tested with the chi-square test. After comparing our data to the lab book data, we came to the conclusion to accept the null hypothesis. Introduction The Sordaria fimicola fungus is commonly found in decaying organic compounds such as in animal or human wastes. Also the fungus has been used to study cross-over because of its ability to show actual characteristics of both the normal and mutant strains of the Sordaria. (Olympus) The wild-type (normal) strain has a genotype of (g+ and t+), which can be identified as the black spores on the asci. The mutant strains have genotypes of (t- and g-) for gray spores, and genotype of (t- and g+) for tan spores, these are usually the lighter ones on the asci. The purpose of the experiment was to understand the rate of Sordaria fungus cross-over through hybridization of between the wild-type and mutant strain. Using the information gathered from the cross-over, calculate the frequency of cross-over to create a map-unit that shows the distance between the genes. We did this lab to see the frequency of cross-over that would occur when normal strains and two mutant strains of the Sordaria fimicola fungi are placed in a nutrient rich Petri dish.
