In translation, the first step in "initiation" is when the small ribosomal subunit binds to the ribosome recognition site on the mRNA. the first amino acid of any newly-synthesized polypeptide is always methionine, because methionine is specified by the 'start'  codon in the mRNA. synthesis of the polypeptide begins with the very first mRNA nucleotide on the 5' end. the first tRNA does not enter the ribosome until after the large subunit has joined the small subunit.
