Permealization helps get the antibody into the fixed cells. Incubation of formaldehy in PBS cross links proteins and other molecules in the cells. Then, incubation of 0.1% sodium AZide in PBS to stop action of formaldehyde, Finally, incubation with 0.5% TITRON X-100 in PBS to permealyze cell membranes bt solybilizing lipids. Why do structures that appear fain the compound light microscope appear strongly visible with a fluorescence microscope? Because the fluorescence microscope illuminates the object with a high energy light, emiting a light that has a lower frequency. With the dark back structure that appear to faint Fluorescence molecules (fluorophores or fluorochromes) absorb light when exposed to the appropriate excitation wavelength.
