DNA Structure The substance of heredity Experiment 1: Griffith infects mice with Streptococcus pneumonia 1928 Injected mice with smooth strain of S. pneumonia (has a silky coat of polysaccharide) The mice died He then isolated the bacteria from those mice and grew them in colonies Colonies had slick appearances (were also smooth); was able to isolate smooth S. pneumonia Injected rough strain of S. pneumonia; mice did not die Without smooth coat of polysaccharide, mice were able to develop a cure against it Since the bacteria did not affect the mice, he could not cultivate any rough strain Injected dead smooth cells into mice and mice did not die Could not cultivate bacteria from mice Injected rough cells with dead smooth cells into mice which became lethal In order for live rough cells to have smooth offspring is for rough cells to BECOME smooth itself; how did that happen? Rough cells changed into smooth cells because of something in dead smooth cells because there was something in dead smooth cells (which caused them to be smooth) that was influencing the rough substance. Saw that the transforming principle looked like Mendel’s genes in that heritability – could be passed from one gene to the next Experiment 2: Avery, MacLeod, & McCarty isolate & characterize the transforming principle 1941 If they could figure out what the transforming principle was made out of – find out what genetic material was made out of Wanted to fractionate dead smooth cells and then isolated polyssacharide fraction, lipid fraction , protein
