5. The process of DNA replication contains many mechanisms for insuring fidelity of information. Both DNA polymerase III and DNA polymerase I have a proofreading ability. When there is a mismatched base, the holoenzyme backs up and removes the nucleotide from the end of the strand. Mismatches trigger exonuclease activity from DNA polymerase I. Once the mismatched base is replaced, ligase then seals the nicks in the DNA 6. The leading strand is synthesized in the 5’ to 3’ direction. This is accomplished in a continuous manner because DNA Polymerase III is synthesizing DNA in the direction of the replication fork’s movement. The daughter strand is formed using free OH groups donated from the RNA primer. The lagging strand differs because it is synthesized in short portions known as Okazaki framents. It cannot be synthesized continuously because the polymerase will not synthesize in the 3’
