Jin Bae Homework #3 1. After the inhalation of Bacillus anthracis, which leads to infection, it is very important to identify the inhibitors of anthrax lethal factor to treat it with antibiotics in its early stage. The purpose of this experiment was to identify the small molecule inhibitors of anthrax lethal factor by running numerous experiments. 2. First experiment: High-throughput screening and hit validation by using HPLC- based assay. The purpose of this experiment was to screen and identify compounds that inhibit LF activity. The experiment was carried out in this manner. After the LF cleaves an optimized peptide, the fluorescence increased. After the standardization of the high-throughput assay, the 1,990 compounds in the NCL diversity set were tested; then more than 75% inhibition were selected. Finally, by using an HPLC-based assay, more than 50% inhibitions were selected. Second experiment: This experiment involved identifying the structure of anthrax LF inhibitors.
