Introduction Mitochondria are considered the primary target of acetaminophen toxicity. 1 The drug attacks the permeability transition. 2 There is a possibility that reactive oxygen species such as nitric oxide and superoxide ions are an important determinate of hepatocyte death. It is thought that non-parenchymal cells that secrete cytokines and chemokines during acetaminophen use induce liver injury. Acetaminophen treatment in mice altered 332 genes including genes involving a stress response, cell cycle, growth inhibition, inflammation and cell signaling. Primary proteomic examinations on acetaminophen- induced injury have honed in on changes in liver proteins expressed in mice, identifying known targets for changes in mitochondrial proteins, heat shock proteins, and other structural and intermediate metabolism proteins. 3 Materials and Methods Chemical materials used included acetaminophen and sodium carboxymethylcellulose, monoclonal anti-actin, polyclonal antibody to aminopeptidase A, ELISA kits, a catalase activity assay kit, rat antibody arrays, bovine serum albumin, beta glycerylphosphate, Angiotensin II, ACTH, and a protease inhibitor. The animals used in this experiment were Male F344/N rats between 10-12 weeks of age and 250-275g. Rats were few with irradiated NTP2000. The rats were then fasted for 14 hours, water was always available. Rats were given as dose of acetaminophen, 10ml/kg and the rats were allowed to feed. The selections of doses and exposure times were calibrated to characterize the serum proteome at early, fulminant and recovery stages of liver injury based on previously published work. The Rats were euthanized by carbon dioxide inhalation and 3-5mL of whole blood was obtained from the inferior vena cava of each rat. No hemolysis was
