The Role of Uncoupling Proteins in Obesity 1. Thermogenin is the uncoupling of protein UCP1. This protein is used to generate heat by non-shivering thermogenesis. UCP1 provides a pathway in which protons can reenter the mitochondrial matrix, linking the respiratory chain to ATP synthase, generating ATP for the cell and therefore allowing respiration and heat production to proceed during the absence of ATP synthesis. 2. a. DNP uncouples oxidative phosphorylation by carrying protons across the mitochondrial membrane, leading to the rapid consumption of energy without the generation of ATP. In a pH gradient, DNP binds protons on the acidic side of the membrane, diffuses through the membrane and releases the protons on the membrane’s alkaline side, thereby acting as a proton-transporting ionophore. The presence of DNP increases its permeability to H + uncouples oxidative phosphorylation from e- transport by providing a route for the dissipation of the proton electrochemical gradient that does not
