Pure culture : culture containing only one kind of microorganism Always use aseptic technique. Two DiFFerent Methods : 1. Streak plate four streak techniques; requires skill and practice a. Quadrant streak we use a modiFed form of this method b. Radiant streak c. T streak d. Continuous streak 2. Pour plate use plates and agar tubes to isolate loopfuls of bacteria; requires less skill and practice but a lot of media and supplies Incubate plates with agar-side up to prevent condensation which may cause the organisms to run together and defeat the purpose of the isolation lame loop and get bacteria lame loop between each new section of streaking Organisms: 1. Serratia marcescens when grown at 25-30 o C, it produces a red pigment called prodigiosin . When grown at higher temperatures, it appears white on the media.When grown at higher temperatures, it appears white on the media....
