Read online guidelines posted by Prof. Yaffe and come to the review session (Wed 7-8 pm, 34-101) Today you will receive a package of problems; we’ll solve some in class, and the rest are for you to prepare for the exam. Solutions will be available during office hours. We will also solve problem set #2. My office hours this week will be on Thursday (2/28) from 5-6 pm, 68-429. Overview of theory + Practice problems Case studies of enzyme catalysis: Chymotrypsin Enzyme as catalysts – classes of enzymes Enzyme Kinetics – find attached another TA’s handout - Michaelis-Menton equation derivation - Lineweaver-Burk plots - Types of inhibitors: competitive, non-competitive, uncompetitive, mixed DNA and RNA – just basic knowledge P1) From the plot of velocity vs substrate concentration written on the board, obtain the following parameters (the amount of enzyme in the mixture is 10 -3 μ m): a) Km b) Vmax c) K 2 /Km d) Turnover number P2) What is the significance of Kcat/Km? P3) Which of the following statement is correct? The turnover for chymotrypsin is 100 s -1 , and for DNA polymerase is 15 s -1 . This means that: A) chymotrypsin binds its substrate with higher affinity than does DNA polymerase B) the velocity of the chymotrypsin reaction is always greater than that of DNA polymerase reaction C) the velocity of the chymotrypsin reaction at a particular enzyme concentration and saturating substrate levels is lower than that of the DNA polymerase reaction under the same velocity conditions
