Andrew Wright Bios 42 Section 66 February 20, 2008 The Cell Nucleus and Packaging of DNA Labs Materials and Methods The Cell Nucleus: Peroxidase Activity and Tetrazolium Reduction in Roots First, place one mL of Tetrazolium, Peroxidase Substrate, and HCl in three respective tubes. Then cut off three 1.5 cm pieces of root tip. Place one root tip into each of the three tubes. Observe the root tips in tubes 1 and 2 for approximately 15-20 minutes until color appears. When it does, remove the roots and rinse them with water. Measure how far the color is located from the tip of the root with a ruler. Microscopic Analysis of Onion Toot Cells First, place the HCl treated root tip from earlier on a microscope slide. Cut off the tip of the root and move it so it is about one inch from the left side of the slide. Cut off a similarly sized piece of the root from the zone of elongation and place it on the right side of the slide. Take a second slide and squish the two slides together with the root pieces facing in. Separate the slides and allow them to dry for 5 minutes. After the slides dry, place a few drops of ethyl alcohol on the cells o both slides. Wait another 3 minutes, then
