Andrew Wright April 3, 2008 Intro to Cellular and Molecular Biology Lab Results and Discussion Enzyme Kinetics Factors Affecting Enzyme Activity Results For this lab, each of the four groups performed Experiments A, B, and C with Groups 1 and 2 doing Experiment D and Groups 3 and 4 doing Experiment E. Experiment A: Effect of enzyme concentration on reaction rate This experiment measured the rate of reaction for varying concentrations of enzymes. The results were measured by recording the absorption of light at 420 nm. Experiment B: Effect of temperature on enzyme activity In this experiment, every group made the same tube. The purpose was, after finding the optimal concentration (.3ml of culture) of enzyme in Experiment A, to see how temperature affected the enzyme activity. The absorbance was measured after the tubes spent 5 minutes in a 90 degree Celsius bath. The reaction was allowed to proceed for 5 minutes before measurement. Experiment C: Enzyme Activity in a culture that has not been induced In Experiment C, enzyme activity was measured using uninduced cultures. Each group measured two different concentrations. Again, the reaction was allowed to proceed for 5 minutes before optical density was measured.
