Daisy Torla Sam Bridges Eric Mangol Noga S Results and Discussion The following figure shows the agarose gel illustrating the plasmids cut by two different restriction enzymes (EcoRI and HinDIII). In lane one the digested pBAG plasmid has been cut with EcoRI. It shows one line of DNA at about 3.4 kilobases. In lane 2 is the digested pBAG enzyme that has been cut with the enzyme HinDIII. There are two lines in this lane. The lower one is at about 3.4 kilobases, while the higher one is at about 12.0 kilobases. The higher line is supercoiled DNA making it difficult to pass through the gel matrix. Lane three has the pBAG plasmid that has been cut with both EcoRI and HinDIII. This lane should have two lines, but it only has one, at about 2.5 kilobases. The missing line would only be about 12 base pairs long, and its diminutive size means it ran off of the gel very quickly before the gel finished running altogether. Lane 4 has the protein standard, which serves as a reference for the kilobase markers.
