BIO325 – February 19, 2008 Mutation (cont.) Bacteria I. Mutation (cont.) a. Nonsense Mutations i. Forward v. Reversion 1. Which is the wild type and which is mutant 2. Wild type – functional, common 3. Mutant – nonfunctional, rare 4. Forward – Wild-Type 5. Reversion – Changes a mutant sequence into a wild type ii. Reversion is when functionality is restored 1. E.g. from a stop codon into protein encoding 2. E.g. transcription rate restoring iii. Reversion – Insertion/Deletion 1. E.g. insertion of single nucleotide in 4 th codon of protein encoding gene 2. Reversion would look like a deletion of the single nucleotide either at the position later 3. Deletion places reading frame back to original status b. Suppression Mutations i. Doesn’t change forward mutation sequence ii. Effect of mutation hidden/suppressed by new mutation iii. In a separate sequence/gene and restores the function that was lost in a previous forward mutation iv. Accounts often enough that we have to account for it v. Observable effects from suppression mutations vi. Not just in bacteria, in other organisms as well (CUT OFF FOR EXAM 1)
