(3 pt)PROCEDURE INFORMATIONMany changes have been made between what you will be doing and thesteps in the lab manual – read this document and know what you willactually be doing.There are many tubes, plates, etc. to use in these experiments. You must besure that you inoculate everything with the right sources and using thecorrect technique.For Ex. 1-4 (aseptic transfer/inoculation):PreparationYou WILL NOT be doing this exercise as written in the manual, although the informationon how the techniques are performed is still valid conceptually and should be read andunderstood.Read pages 31-44 of the manual first. I will provide instruction on doing stab inoculation, butthe technique is done pretty much like it sounds and is essentially a variation of inoculatingbroths.Carefully read the steps for transferring organism from a broth to a broth.Carefully read the steps for transferring organism from a slant to a slant.Think about how these techniques will vary to match the inoculations listed below. Youmay want to create a “cheat sheet” to aid you during lab.The only organism used for this exercise will beEscherichia coli.
Your group will get one set of media (plate, broth, slant) withE. coligrowing on it to use ininoculation. You will share the inoculated media within your group as the source of inoculumfor the cultures you will be preparing.
