C187L/C287L Nano Lab Lab 4 Atomic Force Microscope Experimental procedure for tapping mode Preparation 1. Launch the Nanoscope software on the computer 2. Connect the electrical cable on the AFM head 3. Load your sample into the stage and turn on the vacuum Adjust laser and photo-diode 4. Adjust the laser spot position using the laser alignment skews on top of AFM head such that the laser is incident on the center of the cantilever tip and maximize the sum (~2) as indicated on computer screen. 5. Adjust the photodetector such that the red spot in the meter window moves towards the center of the square, using the detector alignment screws on the left side of the head and set the vertical deflection close to 0 volt. Locate tip and Focus surface 6. Select Stage->Locate tip, center the tip of the cantilever under the cross hairs using the two adjustment screws on the left of head. Focus on the tip end by rolling the trackball while holding down the bottom left button. 7.
