MAE C187/C287L Lab 6. Confocal Microscopy Lab procedure Students who are not familiar with fluorescence microscopy should read the online tutorial, ( http://www.hi.helsinki.fi/amu/AMU%20Cf_tut/cf_tut_part2-1.htm ). Part 1 and part 2 are background. Part 3 is directly relevant to this lab. Power up sequence 1. Turn on mercury (Hg) arc lamp. 2. Turn the Scanner switch on. 3. Power up the Ar laser fan and turn the key to ‘start’ 4. Turn on the GreNe and HeNe laser keys to ‘ON’ 5. Turn on the microscope by orange power switch on lower left side 6. Turn on the computer and start the Leica Confocal Software (LCS) Inverted microscope operation 7. You may choose from a sample of thick plant cells, 3-stain skin fibroblasts, or 2-stain bovine pulmonary artery epithelial cells. Select the 63x objective lens an apply oil on the objective. Mount the stained cell sample facing down on the microscope stage. (Product information is available from Invitrogen, Fluocells products). 8. Focus with the 2 buttons on the right side of the microscope stand and the fine focus knob. Find a region of interest in the sample. Note the theoretical limit of resolution for this lens _________.
