GENE/BIOL3200 Exam II -- Spring 2008 1. (5 pts) Six r mutants of bacteriophage T4 ( a. b, c, d, e, & f ) were each crossed to each other with the following results. Crossing mutants a, c, d, and f to each other gave rise to wild-type recombinants. Mutant b gave rise to wild type recombinants with a and f , but not with c and d while mutant e gave rise to wild-type recombinants with c and d . When mutants b and e were crossed to each other, wild-type recombinants arose. What is the nature of the b and e mutations? Nonoverlapping deletions (b spanning c and d mutants while e spans a and f mutants). 2. (5 pts) Explain how a eukaryotic insulator functions. Why is this an important feature of eukaryotic gene regulation? Use diagrams if you want. A insulator acts to prevent enhancers for one gene acting on neighboring genes. This is important because enhancers can act over very long distances and also allows for their gene specificity (+1 bonus). 5. (10 pts) A DNA sequencing gel using the Sanger dideoxy method is shown below. Read the sequence and write it in the 5'->3' direction. Be sure to label the 5' and 3' ends. 5'- 6. (5 pts) A DNA sequencing gel using the Sanger dideoxy method is shown below. The sequence is also given which reads 5'->3' from left to right. The top of each lane is numbered. Designate which lane represents which dideoxy-NTP reaction (e.g., ddA, ddC, etc.).
