BIO 480W Dr. Gaillard Spring 2009 Protocol for Detection of DIG-Labeled Probe ** Use this protocol after steps 1-5 have been performed on page 113 of your lab manual. *** Perform all of the following steps at room temperature. 1) Place the membrane in a plastic dish and rinse the membrane briefly (1-5 minutes) in Washing Buffer (use just enough to cover the membrane). 2) Incubate the membrane for 30 minutes in 20 ml of Blocking Solution. 3) Incubate the membrane for 30 minutes in 10 ml of Antibody Solution using a Seal-a- Meal bag. 4) Return the membrane to a plastic container and wash the membrane for 15 minutes in 30 ml of Washing Buffer. 5) Repeat step 4.
