The authors want to identify the factors and mechanism of the translocation of GBP from the vegetal shear zone to the future dorsal side. 2. GSK3 phosphorylates β -catenin, which targets it for degradation. After GBP is translocated to the future dorsal side, it inhibits GSK3 activity by binding to GSK3 so that GSK3 dissociates from the protein complex containing Axin, APC, and β -catenin. GSK3 can no longer phosphorylate β -catenin, thus letting nuclear β -catenin accumulate at the future dorsal side. β -catenin then activates the expression of the dorsal organizer genes during zygotic transcription. 3. Using time-lapse confocal microscopy, the authors saw the localization of GBP-GFP in the vegetal region during cortical rotation. GBP-GFP particles showed fast, unidirectional movement in the opposite direction of yolk movement, away from the vegetal pole of the egg. 4. Immunoprecipitation is the technique that separates a protein of interest from other proteins by binding the protein of interest with an antibody specific to that protein. An insoluble antibody-binding protein is added to make the antibody-protein complex insoluble. Centrifugation of solution removes all other proteins.
