PreLab Write-up (BLUE FONTS ONLY) 1.Title: Preparation of a Buffer and pH Measurement 2. Introduction: Water is the universal solvent; water is also an amphoteric substance. It is for this reason that biochemist’s study the properties of water because the addition and removal of H ions causes a change in the pH level of a given solution. Many biochemists carry out reactions that must be performed at a constant pH, it is for this reason that we utilize buffers in order to maintain a desired pH. A buffer is defined as a solution containing a mixture of a weak acid [HA] and its conjugate base [A-] which is capable of resisting substantial changes in the pH following addition of acidic or basic molecules. The pH may be defined at the –log of the Hydrogen Ion Concentration, it can be calculated several ways one of which is by taking the –log of the given Hydrogen Ion concentration. A thorough understanding of how acids and bases affect pH is mandatory for biochemistry.
