BIOCHEMISTRY NEED TO KNOW Chapter 11 Chapters 11 and 12 will unite the concepts and molecules learned in all the rest of the course thus far. The general theme is the structure and function of Biological Membranes. The concepts and models presented in these chapters are very important for an understanding of the Biochemical pathways that will be mastered in the second module of this course, and for Cell Biology courses later in your academic program. Membranes are composed of proteins, carbohydrates and lipids. An understanding of membrane function is derived from understanding the various roles of lipids and proteins in membrane structure. A major theme running through all this material is this: membrane function is entirely dependent upon the number and kinds (and the properties of) of proteins and lipids that comprise them. Altering the concentration of phospholipids or of a specific membrane protein can and will lead to either enhanced or restricted membrane function. Finally, the environment on either side of membranes plays a very important role in membrane function, as you shall see. Membrane lipids : Understand the three basic structures of lipids in aqueous environments (figure 11-4) and why liposomes are the most stable form for cells to use. Note that they provide for two different aqueous environments because of their structure. Understand that lipids are asymmetrically distributed in membranes and differently distributed in different cells and organelles (see figures 11-2 and Table 11-1, for example). Membrane Proteins : Understand the classification: Peripheral versus Integral proteins, Transmembrane proteins, and lipid-linked (figure 11-14) membrane proteins Understand the classification of integral membrane proteins (figure 11-8) Understand the Hydropathy Index Membrane Carbohydrate: Understand the orientation of carbohydrates in the plasma membrane Understand that carbohydrates are found on intracellular membranes and on what membranes you might find them. 1

Understand the basic course of events that comprise membrane fusion (figures 11-23 and 11- 24, for example). Membrane transport mechanisms are central to cellular maintenance. Chapters 11 and 12 offer many examples of membrane proteins, membrane transport and membrane function. It is more important to understand the mechanisms that membranes use to get solute into and out of cells that to memorize all the little details of each system (but an understanding of some detail is needed). As you go through examples, keep in mind why they are in a section of the chapter, what features make them function in the role as presented in that part of the chapter, and what controls exist to limit transport. You should be able to look at an example that is not in the text and be able to identify the kind of mechanism required to activate it (such as ligand-gated).
