DNA EXTRACTION FROM A BANANAIntroduction:Deoxyribonucleic acid (DNA) is a nucleic acidusually in the form of a doublehelixthat contains the genetic instructions monitoring the biological development ofall cellular forms of life, and many viruses.A complete copy of DNA is found in every cellin any organism. In order to release the DNA to analyze it, scientists must break openthe cells and remove structural proteins and enzymes that interfere with the DNAstructure. This simplified procedure releases a great deal of DNA so that you can see it.It allows observation of DNA’s physical and chemical properties.Materials:NaCl (table salt)meat tenderizerWoolite detergentelectronic balance or food scaleBanana100ml beaker or cupa scientific thermometerhot water bath (pot of water at60°C)coffee filterfunnelIce bathice-cold ethanolProcedure:1.Prepare the DNA extraction solution in a 250ml beaker:a.Dissolve 2g of NaCl (table salt) in 90ml of H2Ob.Add a sprinkle of meat tenderizerc.Add 10ml of Woolite detergentd.Stir gently trying not to create bubbles2.Using an electronic balance and weighting paper weight out 30g of banana.Besure to set the balance to grams and ‘zero’ it before weighting. If you areunclear about how to do this make sure to ask your instructor3.Mash the banana until smooth.4.Once the banana is smooth transfer it to a 100ml beaker or cup.Add theextraction fluid until the total volume of solution and fruit is twice that of thefruit alone and gently stir. Note: your solution must be liquid, if it is too viscous(thick), add more extraction solution.
