Mutagenesis of Saccharomyces cerevisiae Materials required per group: Sterile Microfuge tubes Microfuge tube rack Sterile 200-1000 µl pipet tips Sterile 20-200 µl pipet tips 20ml of sterile water in a test tube. 8 YPD plates. 6 Sterile plate spreaders. Sterile toothpicks Test tube rack Sharpie Items required per class: Sterile culture tubes Stratalinker 1800 200 ml YPD S. cerevisiae strain HAO S. cerevisiae strain HBO Spectronic 20 and cuvettes Petroff-Hauser chamber or 0.1mm hemocytometer 28°C rotatory shaker 28°C incubator 37°C incubator Bunsen burner Sterile 25 ml pipettes 1. Prepare 10ml ON cultures of HAO and HBO in YPD at 28°C two days before the first day of class. 2. Inoculate a 500ml flask, containing 100ml YPD, with 100µl of HAO and a second flask with HBO. Grow at 28°C with constant shaking for around 8h. DAY1 3. Determine the titer of the cultures using a hemocytometer for the whole class. 4. Transfer 5ml of each culture (HAO or HBO) into cuvettes and measure the
