Chromosome Mapping (Three-Point Test Cross) Objective The objective of this experiment is to determine the linkage distances between three genes that are located on the X-chromosome. Variables Independent -Diet for the flies (food supply) -Phenotype and genotype of males used in test cross (wild) -Length of experiment Dependent -Genotype of virgin females used ( w m f ) Hypotheses H 0 : There will be no significant difference between the expected mapping distance and the experimental mapping distance of the X-linked genes. H 1 : There will be a significant difference between the expected mapping distance and the experimental mapping distance of the X-linked genes. Conclusions I reject my null hypothesis.

Introduction The objective of this experiment was to determine the linkage between three genes located on the X chromosome. All of the genes tested were recessive, X-linked genes. We expect that there will be a significant difference between the expected and the experimental mapping distances. The independent variables in this experiment were the length of the experiment, the males used in the test cross, and the chosen food supply. The dependent variable was the genotype of the virgin female flies used. Materials 1. Male flies (Wild phenotype) 2. Virgin female flies ( w m f ) 3. cotton to cap the food bottles 4. Fly food for the food bottles 5. Yeast for the adult fly diet 6. Food bottles 7. Antifungal paper to place in food bottles 8. Funnel to transfer and count flies 9. Fly nap to aid in transfer and counting Methods This experiment was set up on March 6, 2008. It ran for five weeks and was completed on April 10, 2008. We used only 2 bottles of flies due to shortage of female
