MEASURING PROTEIN SOLUTION Abstract In the third lab, Measuring Protein Solutions, explains how to quantitate the amount of protein in a solution. Then, determine if the Biuret assay can detect both proteins and amino acids. Next, follow all the procedures as instructed to do so in the lab manual, Biol1205 Cells and Molecules Laboratory. Also the materials that will be used included a LabQuest, SpectroVis Plus, cuvettes, vortex, as well as test tubes, pipettes, and all the solutions that are also listed in the lab manual, Biol1205 Cells and Molecules Laboratory. In conclusion, this lab will improve the understanding of how to mix solutions, and then be able to measure the amount of proteins. 2

MEASURING PROTEIN SOLUTION Introduction The purpose of this lab is to determine the amount of proteins in a solution. But in order to get the correct measurements of proteins in each test tube we have to salt them out. The salt helps to show the proteins because without the proper amount of salt the proteins will not show up in our SpectroVis. During our experiment we did not add enough of salt into each test tube that was supposed to have it. We then realized it, it was too late, but we learned how valuable the salt truly is when trying to make the proteins as visible as they were. Because there was not enough of salt in our solutions, the results that we received were too low to be even considerably accurate. So we then collaborated with our classmate who’s salt content was the proper amount to truly understand how much salt was needed and how that really affected our results. After we added our salt, in order to use the SpectroVis we had to vortex our test tubes for ten minutes that was the test tube will be properly diluted. We vortex each test tube by putting in the centrifuge.
