4 How do we define a gene? The minimal segment of a chromosome that can cause a phenotype Complementation test • Also known as the cis-trans test – Cis=same locus – Trans=different locus Test for ‘rescue’ (complementation) of a phenotype Non-complementation : If two mutations lie in the SAME locus, then they are allelic . that is they are different alleles of the same gene. Complementation : If two mutations lie at DIFFERENT loci, then they are nonallelic that is they are different genes •Rescue means that a wildtype copy of each phenotype must be inherited •Phenotypes must be inherited in a recessive manner
