(20 pts) The pBR322 and pUC19 plasmids are used as a vectors in recombinant DNA procedures to clone genes. pBR322 carries tetracycline and ampicillin resistance genes used for selection, whereas pUC19 carries an ampicillin resistance gene and a LacZ’ gene for selection. In independent experiments , a gene coding for human lactoferrin was inserted into a restriction site in the located inside the ampicillin resistance gene in pBR322, and in the LacZ gene in pUC19. Then the plasmids were then introduced by transformation into E. coli cells. Describe the procedures you will follow in each case to select the bacterial colonies carrying the recombinant plasmids, including media composition. pBR322: Growth the bacteria after transformation in a master plate containing antibiotic free medium. Replicate plate in 2 plates. The first one containing tetraciclin to select for transformed bacteria, and the second one containing both ampicillin and tetracycline. Colonies containing recombinant DNA plasmids will not grow in the plate since the ampicillin resistant gene has been disrupted by the introduction of the lactoferrin gene. Therefore these can be selected from the master plate. pUC19: Grow bacteria after transformation in a plate containing ampicillin and the lactose analog such as x-gal/IPTG that turns blue when the LacZ’ gene is functional.
