Microbiology Chapter 7 – Essay Questions 1.Describe various types of nucleic acids that are typically found in cells. (Section: 2. Bacterial strain A contains a plasmid. Bacterial strain B does not. When the bacteria are incubated together in a broth culture, strain B cells containing the plasmid can be isolated. Devise an experiment to determine what type of gene transfer process is involved. (Section: Genetic Recombination and Transfer, Learning Outcome: 7.28). This type of gene transfer is bacterial conjugation. Bacterial strain A is considered to be an F plasmid cell also known as F+ cells, this will serve as a donor to Bacterial strain B which is known as the recipient cells (F- cells). Both bacterial strains are incubated together in the broth culture. A sex pilus connects a donor cell (F+) which is Bacterial strain A to the recipient cell (F-) which is Bacterial strain B. plasmid, takes one strand of the plasmid DNA and this single strand transfers into the recipient, starting with a section called origin of transfer. Both cells synthesize complementary strands of F plasmid DNA to form complete double stranded DNA circles. Now Bacterial strain B (the recipient cell) is now an F+ cell (containing a plasmid). 3. Compare and contrast the lactose operon with the tryptophan operon. (Section: Gene Function, Learning Outcome: 7.17) Lactose operon – is an inducible operon and the first operon whose structure and action were explained.
