Lecture 6 10/10/07 Nuclear Import/Export Nuclear Import Receptors – Importins Helps cargo get into nucleus – does this by binding to cargo (protein with NLS) and bringing it into nucleus by mediating nuclear import Three kinds: 1.) Importin – specific for: o Bipartite NLS o Classical NLS 2.) Transportin – specific for M9 NLS 3.) Snurportin – snRNP (“snurps”) – small ribonucleo proteins, or proteins involved in RNA splicing; specific to a kind of RNA binding protein FG Repeats – Amino acid sequence of nucleoporins Make up cytoplasmic filaments Importins bind to repeats to get through nuclear pores Importins – 2 kinds α and β (work together) 1.) Alpha and beta importins bind cargo + NLS 2.) Alpha and beta importins mediate transport via FG repeats of NUPs 3.) RAN–GTP binds beta importin dissociates from cargo, accompanies importins out of the nucleus 4.) RAN–GTP hydrolyzed to RAN–GDP via GAP in cytoplasm provide energy for transport 5.) RAN separates from importin alpha and beta when it gets hydrolyzed
