Prac 3: Amylase Activity in Germinating Barley 1. You have performed an experiment (your practical 3, Metabolism experiment). You wish to tell your scientist friend about this experiment so they can replicate it exactly. Write a methods section for this practical. Pat dry 10 germinating seeds and record the weight of the germinating seeds. Crush the 10 germinating seeds into a paste. Further crush the germinating seeds for 3 minutes after adding 10ml of buffer to allow for the amylase to go into the solution. Filter the crushed seeds and measure the volume of the amylase solution. A five to one dilation is done with 5ml of amylase extract and 20ml of buffer is added to make the diluted amylase extract. A control with 5ml of diluted amylase extract in an 80°C water bath for 10 minutes. The purpose of the control is used to compare when the independent variable is changed ( Crowder, 2013 ). The high temperature denatures the amylase in the control and therefore it can’t react to the starch and produce sugar. The control, boiled amylase extract did not reach achromic point because the enzyme had denatured due to heating. If achromic point was reached with the control experiment, this suggests that the control tube was not placed in the water bath soon enough that reactions had occurred.
