CELL RESPIRATION LAB PURPOSE PROCEDURE 1. Set up a room temperature bath (approximately 25 °C) and a 10 °C bath. Add ice to attain 10 °C. 2. Respirometer 1: Fill a 100 ml graduated cylinder with 50 ml of water. Put in 25 germinating peas and measure the amount of water displaced (volume of peas). Record the volume of 25 germinating peas. Dry peas and use them for respirometer 1. Respirometer 2: Fill the graduated cylinder with 50 ml of water. Put in 25 dried peas into the cylinder and then add enough glass beads to obtain a volume equivalent to that of the germinating peas. Dry peas and beads and use them for respirometer 2. Respirometer 3: Fill the graduated cylinder with 50 ml of water. Put in as many glass beads that are needed to obtain a volume equivalent to that of the germinating peas. Remove these beads and place them on a paper towel. They will be used in respirometer 3. 3. Make the respirometers by placing a small wad of absorbent cotton in the bottom of a vial and saturating the cotton with 15% KOH (potassium hydroxide). Then, place a small wad of dry cotton on top of the KOH – soaked absorbent as shown in the apparatus diagram at the end of this text. The amounts of cotton and KOH should be the same. Put the germinating peas, dry peas + beads, and beads in vials 1, 2, and 3, respectively 4.
