Study Guide Exam 3 Here are the important concepts from each lecture that I think are worth knowing. If something is worth knowing, it may appear on the test. I cannot test on everything that I want you to know (unless of course you want me to write a 3 hour exam!). However, if you can answer the following questions, you will do very well on the 3 rd exam. Regulation of Metabolism - about 14 questions 1. Explain the regulatory circuit for each of the following systems. In addition be able to predict what inactivation of each of the genes in the operon would do: 1. The lac operon 2. The trp operon 3. The heat shock genes 4. Quorum sensing in Gram-positive ( S. aureus ) and Gram-negative bacteria ( V. fischeri ) 5. Know what riboswitches and know how they can regulate DNA Genomics - about 12 questions 1. Describe the difference between a plasmid and a chromosome. 2. How are plasmids maintained in the cell? Describe several maintenance systems that have been discovered. 3. Describe the 3 methods of horizontal gene transfer that have been discovered in bacteria. Which one uses a virus? Which one involves the uptake of naked DNA. Which one involves the use of a sex pilus? 4. Explain the difference between a selection and a screen. 5. Define a silent mutation, nonsense mutation, point mutation, frame-shift mutation, and deletion. 6. List the types of modifications to the DNA that lead to mutation, explain how they occur, and how the cell repairs them.
