AtLee Watson Experiment 9: Column Chromatography: Separation of Nitroacetanilides In this experiment column chromatography was used to purify the crude nitroacetanilide product from Experiment 8, derived from the nitration of 4-methylacetanilide. To prepare the column, it was first filled halfway with hexanes:ethyl acetate (solvent in 2:1 ratio) and then packed with alumina, the solid adsorbent. After a layer of sand was added and 10 mL of solvent was eluted through the column, the dissolved product was poured on top and eluted through the column. Purified product was collected as the yellow bands approached the bottom. After all of the product was collected it was combined and was evaporated to give the solid product. From an initial amount of 0.97g, 0.68g was recovered yielding a 70% recovery. Recrystallization and analysis of the product using TLC chromatography was not required in this experiment. 1. The fractions which contained the desired compound were yellowish in color.
