Study Guide MIMG 100L Week 1 Exercises: Pure culture =a single species on a medium Mixed culture =two or more species on a medium Agar slants =generally used to grow stock cultures that can be refrigerated after incubation and maintained for several weeks Plated media =used for obtaining isolation of species, differential testing, and quantifying bacterial densities Streak plate = a commonly used isolation technique to obtain pure cultures from a mixed culture. During streaking the cell density decreases, eventually leading to individual cells being deposited separately on the agar surface. Cells that have been sufficiently isolated will grow into colonies consisting only of the original cell type. · Quadrant streak=used with samples suspected of high cell density o Beginning the quadrant streak pattern=streak the mixed culture back and forth in one quadrant of the agar plate. Stay close to the plate’s edge and make the streaks long. Don’t cut the agar with the loop. Flame the loop after done. o Second streak=Rotate the plate nearly 90 degrees and touch the agar in an inoculated region to cool the loop. Streak again, using the same wrist motion.
