Quantifying Chlorophyll May 2, 2007 Abstract . The experiment was conducted to determine how chlorophyll content, and chloroplast concentration varies on different sections of the same leaf. The chlorophyll content and the chloroplast concentration were found to be the greatest in the middle section of the leaf. The chlorophyll content in the middle sample was 296.6 mg/g and the chloroplast concentration was 4.4x10 6 chloroplasts/ml. Introduction The chlorophyll content of leaves is dependent of many factors which include nitrogen availability, light, physiological stress, and senescence. The element nitrogen is a component of the chlorophyll molecule, so the amount of nitrogen available will lead to higher chlorophyll content. Light, which starts the synthesis of chlorophyll, is also a factor that will lead to an increase in chlorophyll content. An exception to this rule is found in spinach leaves, where an amplified amount of light will not necessarily lead to a rise in chlorophyll content. Spinach leaves will be used in this experiment. Chlorophyll content will also vary depending on a leaf’s location on the stem of the plant; leaves in the middle usually have greater chlorophyll content than leaves of the top and bottom. Chlorophyll is contained within chloroplasts (Kaisa, 2008). Chloroplasts are the site of photosynthesis, the process in which light energy is converted to chemical energy (Mader, 2005).
