Eiman Henson Biology 101L Hills Procedure Outline I. Procedure A. Step 1 1. Spectrophotometer will be set to 605 nm. 2. Don’t move wavelength dial 3. Prepare suspension of chloroplasts from leaves 4. Record data on data chart B. Step 2 1. Make sure you have all materials (test tube rack, goose-neck light, aluminum foil, 4 spectrophotometer tubes, ice container with ice, 1 mL transfer pipettes, Kimwipes, distilled water, 0.1-M phosphate buffer, Parafilm, large Erlenmeyer flask filled with water C. Step 3 1. Number cuvettes 1-4, respectively 2. Use Kimwipes to wipe outside of each cuvette clean 3. Handle carefully

D. Step 4 1. Make foil cover and cap for dark tube so that no light can enter covette 2. You will need to remove foil when inserting tube in spectrophotometer at each reading E. Step 5 1. Obtain suspension of boiled and unboiled chloroplasts from instructor 2. Keep them on ice F. Step 6 1. Add buffer, water, and DPIP to appropriate cuvettes (1-4) and use chart to do this 2.
