2 DNA Replication As cells pass through the cell cycle, the DNA must be copied, or replicated , before mitosis and cytokinesis. DNA is replicated during S (DNA synthesis) phase of the cell cycle. When the structure of DNA was first worked out, its double helical structure immediately suggested how replication may occur: each of the two strands would be copied.
