September 18 TH DNA Replication -Semiconservative: one of the 22 strands is conserved in each progeny molecule -Bidirectional: it proceeds in both directions from the same starting point (origin of replication) E.coli chromosome is circular AT has one H bond therefore easier to break The replication fork (helicase unwinds the double helix): Need - 2 types of DNA polymerases- enzymes that catalyze addition if nucleotides to a growing polynucleotide DNA chain -DNAP1, adds nucleotides in a 5’ to 3’ direction -DNAP2, adds nucleotides in a 3’ to 5’ direction DNA replication: -starts when helicase unwinds the double helix into a replication fork; strand separation -Primase: initiates replication by making a short RNA primer: puts a ribonucleic acid primer (RNA) to start replication -DNAP: adds nucleotides to the 3’ OH groups of growing DNA strand ( with RNA primer as starter) - binds to the primer and synthesises the new strand – can only synthesis in
