Title : Determining the effect of substrate concentration on the rate of catalase enzyme reac- tion Introduction: Catalyze is a common enzyme found in most living organisms. The function of a catalase is the decomposition of Hydrogen Peroxide into water and oxygen. Equation: 2H 2 O + catalase-------> 2H 2 O + O 2 In this experiment, I will attempt to investigate the effect of substrate concentration of the rate of enzyme catalase. The substrate I will be using is Hydrogen Peroxide (H2O2) with different lev- els of concentration. I will dip filter paper discs into the yeast solution for 3 seconds and then drop it into beakers with different H2O2 concentrations (0%, 1%, 2% ,3% ,4% and 5%). I will then time the amount of time it takes for the filter paper to rise back up. The general idea is, the faster the disc rises, the faster the enzyme is working therefore more oxygen is produced. Aim: To investigate the effect of substrate concentration on the rate of enzyme catalase Hypothesis: My hypothesis is that as the concentration of hydrogen peroxide increases, the rate of reaction will increase. This is because molecular collisions as in higher concentrations there are more movements of enzymes and there is a higher possibility of the enzyme fitting into an active site. Consequently, the rate of reaction will decrease at low concentration as there will be fewer colli- sions between the substrate and the enzyme molecules. This will results to lower production of oxygen. Therefore, I predict the rate of enzyme catalase is most efficient at 4%, and 5% sub- strate concentration and least efficient at 1% substrate concentration with 0% concentration having no effect at all. Materials - Hydrogen Peroxide - 2.5 grams of yeast - Beaker (100mL) (x5) - Graduated Cylinder (10mL) - Filter Paper - Hole Puncher - Distilled Water - Forceps
