Hypothesis: In this experiment, we will be testing the effects of pH on the enzyme peroxidase. We want to know at what pH’s the peroxidase will stop working. We expect that pH’s in the extremes will cause the enzyme to denature and no longer work. We expect this because we know that changes in the pH will cause an enzyme to no longer work. We chose to set up the experiment this way in order to test a large variety of pH changes with an easy way to measure the efficiency of the enzyme (using a spectrometer). List of Materials: -Computer with LoggerPro -Spectrometer with Cuvette -Peroxidase -Distilled Water -Turnip Juice -pH Buffers (4, 5, 6, 7, 8, 9, 10) -Pipet -Beakers -Test Tubes -Lab Bench -Timer Methods and Procedure: Prepare for the Experiment 1. Startup LoggerPro. 2. Click on Experiment then Calibrate then Spectrometer. 3. While the lamp is warming up, pipet 2 mL of pH 7 buffer into an empty cuvette. Pipet 0.5 mL turnip extract into this cuvette. Pipet up and down a few times to mix thoroughly.
