Lab 1 Notes Exercise 1. Use of the Bright-Field Light Microscope Microscope Type I. Binocular Compound Microscope -Binocular - Two ocular lens -Compound - Two lens system- includes objective lens and ocular lens -Parfocal - Specimen remains in focus as you navigate low, high, and oil immersion objectives; requires only a slight adjustment of the fine focus. II. Magnification of the Lenses Ocular Lens: 10 X (magnification) Objective Lens: 1) Low-dry 10X (use coarse adjustment) 2) High-dry 40X (use only fine adjustment) 3) Oil-immersion 100X (use only fine adjustment) III. Total Magnification Total Magnification= Objective magnification x Ocular magnification 100X = 10 x 10 400X= 40 x 10 1000X = 100 x 10 IV. Important Concepts to Know! 1. Condenser - focuses light onto specimen. 2. Iris diaphragm - allows you to adjust the amount of light on specimen. 3. Oil - minimizes refraction (bending of light) to enhance resolution when using oil-immersion objective.
