Culture Transfer Techniques Aseptic Technique - organisms are cultured free of contamination. 1) Broths - growth of fresh cultures in large numbers 2) Agar slants - maintenance of cultures 3) Agar deeps - testing for motility and oxygen requirements *Growth at the top of tube- strict aerobe *Growth at the bottom- strict anaerobe *Growth throughout- facultative anaerobe Culture Transfers: (Draw tubes) Broth tube 1. M. luteus (slant) Inoculating Loop Slant tube Broth tube 2. M. luteus (broth) Inoculating Loop Slant tube 3. M. luteus (broth) Deep tube Inoculating Needle M. luteus (slant) Deep tube Inoculating Needle Materials needed by TA’s Rack of Broth tubes (cold room between 210/216) Rack of Slant tubes (cold room between 210/216) Rack of Deep tubes (cold room between 210/216) Bacterial cultures (Micrococcus luteus)- usually put out by Elizabeth or Pat *Note: Will incubate cultures in 37⁰C incubator and later transfer to fridge.
