Introduction:The process of successfully separating an individual microorganism from a mixedpopulation is called isolation. A culture containing many kinds of microbes is knownas a mixed culture. One that contains a single type of microorganism is called a pureculture. As bacteria multiply on or in solid media, visible clumps form cells areformed. Two methods used to attain isolation are the streak plate method ad thepour plate method. The streak plate method consists of aseptically spreading theinoculum over the surface of a sterile agar plate.The large population of cells will begradually reduced so that individual cells are distributed over the agar’s surface.Each cell will produce a colony. This method of isolation is one of the most widelyused techniques in the bacteriology laboratory.1.
