Jason Thomas Monday 9am Lab Period TA: Monica T. Chapter 7 Lab Report Introduction and Purpose The purpose of this lab is to study and examine the different stages of growth in a bacterial sample. Using the bacteria Escherichia Coli students were to study the growth of the bacteria in two different environments, room temperature and 37 degrees Celsius. By using a spectrometer students were able to evaluate when the specimen was experiencing the different phases of bacterial growth, lag phase, log phase, stationary phase and the death phase. Using 2 different types of media, GMS plate and a Luria broth agar plate we can compare the different ways that bacteria grows given the type of media it is inoculated in. Bacterial growth is dependent on several factors which include temperature, pH, osmotic pressure, nutrient availability and type of media. Materials LB tube Eppendorf tubes P200 Micropipette 3 LB plates Rack for Eppendorf tubes Spectrophotometer Ecoli Culture Methods 1) Transfer 1ml culture of Luria broth into cuvette and place in spectrophotometer to zero out the reading.
